Nucleotide sequence of the streptothricin-acetyl-transferase gene sat-2.

The streptothricin resistance (St) gene of the transposon Tnl826 was cloned (1) and sequenced (Fig.) A 534 bp open reading frame could be translated starting with a GTG at position 506 (178 amino acids) or with an ATG at position 518 (174 amino acids). Translation as proposed (Fig.) would result in a polypeptide of predicted size 19670 daltons identical to the C-terminal part of the translation...

Cloning and characterization of the streptothricin-resistance gene which encodes streptothricin acetyltransferase from Streptomyces lavendulae.

The streptothricin-resistance gene of Streptomyces lavendulae No. 1080 was cloned in S. lividans using pIJ41 as a vector. From subcloning experiments, the 1.6 kb BamH I fragment was determined to encode the structural gene. The cell extracts of S. lividans carrying the gene on the plasmid pKS7 had activity to inactivate streptothricin in the presence of S-acetyl coenzyme A, indicating that the ...

Nourseothricin acetyltransferease: a positive selectable marker for Toxoplasma gondii.

Molecular analysis of parasite genomes will require new molecular genetic tools. The nat1 gene of Streptomyces noursei encodes nourseothricin acetyltransferase, conferring resistance to the aminoglycoside antibiotic nourseothricin. Electroporation of nat1 cassettes into RH or Prugniaud strains of Toxoplasma gondii allows for selection of stable nourseothricin-resistant clones.

Localization of Methyl-Coenzyme M Reductase as Metabolic Marker for Diverse Methanogenic Archaea

Methyl-Coenzyme M reductase (MCR) as key enzyme for methanogenesis as well as for anaerobic oxidation of methane represents an important metabolic marker for both processes in microbial biofilms. Here, the potential of MCR-specific polyclonal antibodies as metabolic marker in various methanogenic Archaea is shown. For standard growth conditions in laboratory culture, the cytoplasmic localizatio...

A Versatile Medium for Cultivating Methanogenic Archaea

BACKGROUND Methanobrevibacter smithii, Methanobrevibacter oralis, Methanosphaera stadtmanae, Methanomassilicoccus luminyensis and Methanobrevibacter arboriphilicus have been cultured from human digestive microbiota. Each one of these fastidious methanogenic archaea requires a specific medium for its growth, hampering their routine isolation and the culture. METHODOLOGY/PRINCIPAL FINDINGS A ne...
